以吉林省最大湿地保留区镇赉县为研究区,基于1980—2018年8期土地利用数据,采用网格分析法研究湿地景观格局变化规律及景观指数与人为干扰度的关系。结果表明:(1)镇赉县湿地面积减少290.7 km2,水体、滩地、沼泽变化率分别为-0.45,-2.97和-4.84 km2·a-1。(2)镇赉县湿地率(PLAND)下降6.1%,且从西北部向东南部逐渐递增;斑块数量(NP)与斑块平均面积(AREA_MN)均呈先上升后下降的趋势,东南部湿地景观破碎化加剧,中部破碎化减缓;湿地连通度的空间分布格局为西北向东南递增,斑块结合度指数(COHESION)在中部地区显著增加,在东南部地区缓慢降低。(3)镇赉县湿地人为干扰度逐渐增强、耕地面积的增加和交通网的扩张是镇赉县湿地面积萎缩、景观破碎化加剧和连通度降低的主要人为原因。人为干扰度与NP呈显著正相关,与PLAND、AREA_MN、COHESION均呈负相关。稳定西南部和东北部耕地中湿地斑块,进一步加强中部湿地水体连通性,排查东南部湿地核心区违规建筑,实施分区治理,加强对镇赉县现有湿地的保护,以期实现区域可持续发展。  相似文献

Triapine is an inhibitor of ribonucleotide reductase (RNR). Studies have shown that triapine significantly decreases the activity of RNR and enhanced the radiation‐mediated cytotoxicity in cervical and colon cancer. In this work, we have developed and validated a selective and sensitive LC‐MS/MS method for the determination of triapine in human plasma. In this method, 2‐[(3‐fluoro‐2‐pyridinyl)methylene] hydrazinecarbothioamide (NSC 266749) was used as the internal standard (IS); plasma samples were prepared by deproteinization with acetonitrile; tripaine and the IS were separated on a Waters Xbridge Shield RP18 column (3.5 µm; 2.1 × 50 mm) using a mobile phase containing 25.0% methanol and 75.0% ammonium bicarbonate buffer (10.0 mm , pH 8.50; v/v); column eluate was monitored by positive turbo‐ionspray tandem mass spectrometry; and quantitation of triapine was carried out in multiple‐reaction‐monitoring mode. The method developed had a linear calibration range of 0.250–50.0 ng/mL with correlation coefficient of 0.999 for triapine in human plasma. The IS‐normalized recovery and the IS‐normalized matrix factor of triapine were 101–104% and 0.89–1.05, respectively. The accuracy expressed as percentage error and precision expressed as coefficient of variation were ≤±6 and ≤8%, respectively. This study developed a method for the simultaneous determination of erlotinib and its isomeric major metabolites, OSI‐413 and OSI‐420, in human plasma using an isocratic liquid chromatography–tandem mass spectrometry. Plasma specimens deproteinized with acetonitrile were separated using a 3‐µm particle size octadecylsilyl column. The m/z values of the precursor and product ions for the analytes were as follows: erlotinib, 394.2/278.2; and OSI‐413 and OSI‐420, 380.2/278.2. The total run time was 21 min and no peaks interfering with the analytes and internal standard (d6‐erlotinib) in human plasma were observed. The calibration curves of erlotinib, OSI‐413 and OSI‐420 were linear over the concentration ranges of 10–3000, 2–500 and 2–100 ng/mL, respectively. The pretreatment recovery ratios were >86.1%. The intra‐ and inter‐assay precisions and accuracies were <12.7 and 89.0–108.9% for all analytes. This validated method was applied to the determination of plasma samples in lung cancer patients receiving 150 mg of oral erlotinib. The plasma concentration ranges of erlotinib, OSI‐413 and OSI‐420 were 373–2354, 15.7–379 and 2.5–43.6 ng/mL, respectively. In conclusion, the present method can be helpful for evaluating the plasma exposures of erlotinib and its major isomeric metabolites in clinical settings. Although some polyphenols are known to possess anticancer activity against different cancer cell lines through induction of apoptosis, the mode of antiproliferative effect of ethyl gallate against human oral squamous carcinoma cell line KB was not studied until now. Therefore, the antiproliferative effect of ethyl gallate was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay in comparison with the reference drug paclitaxel. Generation of reactive oxygen species, mitochondrial membrane potential loss, DNA damage and apoptosis were determined using 2,7-diacetyldichlorofluorescein fluorescence, uptake of rhodamine-123 by mitochondria, comet assay and acridine orange/ethidium bromide dual-dye staining method. Both ethyl gallate and paclitaxel exhibited cytotoxicity in a dose-dependent manner. The 50% inhibitory concentration for ethyl gallate was 30 and 20 μg/mL for paclitaxel. A volume of 50 μg/mL of ethyl gallate was found to be significantly effective (P < 0.05) in controlling the cancer cell proliferation leading to acute apoptosis.  相似文献

Water bodies located nearby cities are much prone to pollution, especially in the developing countries, where effluents treatment facilities are generally lacking. The main reason for this phenomenon is the increasing population in the cities, and the large number of industries located near them. This leads to generation of huge amounts of domestic and industrial sewage that is discharged into the water bodies, increasing their organic pollutant load and resulting in the depletion of dissolved oxygen. In this paper, we propose a mathematical model for this situation, focusing especially on the resulting quality of the water, determined by the level of dissolved oxygen. The model also accounts for resources needed for the population survival and for the industrial operations. In addition, we describe also the decomposition of organic pollutants by bacteria in the aquatic medium. Feasibility conditions and stability criteria of the system's equilibria are determined analytically. The results show that human population and industries are relevant influential factors responsible for the increase in organic pollutants and the decrease in dissolved oxygen in the water body, in the sense that they may exert a destabilizing effect on the system. Single nucleotide polymorphisms (SNPs) are the most common genetic polymorphisms and play a major role in many inherited diseases. Methylenetetrahydrofolate dehydrogenase 1 (MTHFD1) is one of the enzymes involved in folate metabolism. In the present study, the functional and structural consequences of nsSNPs of human MTHFD1 gene was analyzed using various computational tools like SIFT, PolyPhen2, PANTHER, PROVEAN, SNAP2, nsSNPAnalyzer, PhD-SNP, SNPs&GO, I-Mutant, MuPro, ConSurf, InterPro, NCBI Conserved Domain Search tool, ModPred, SPARKS-X, RAMPAGE, FT Site and PyMol. Out of 327 nsSNPs form human MTHFD1 gene, total 45 SNPs were predicted as functionally most significant SNPs, among which 17 were highly conserved and functional, 17 were highly conserved and structural residues. Among 45 most significant SNPs, 15 were predicted to be involved in post translational modifications. The p.Gly165Arg may interfere in homodimer interface formation. The p.Asn439Lys and p.Asp445Asn may interfere in binding interactions of MTHFD1 protein with cesium cation and potassium. The two SNPs (p.Asp562Gly and p.Gly637Cys) might interfere in interactions of MTHFD1 with ligand.  相似文献

A high‐throughput miniaturized liquid–liquid extraction procedure followed by a simple ultra‐high performance liquid chromatography method coupled with fluorescence detection for bioanalytical analysis of all tocopherol isomers and retinol in human serum has been developed and validated. In the extraction procedure, a synthetic internal standard tocol was used, which does not occur in the human body. The separation of structurally related vitamins was achieved using a new generation of pentafluorophenyl propyl core–shell stationary phase with elution using methanol and an aqueous solution of ammonium acetate. The fluorescence of retinol and tocopherol isomers was detected at λex = 325, 295 nm and λem = 480, 325 nm, respectively. The rapid baseline separation of all analytes was accomplished within 4.0 min. The sensitivity of method was demonstrated with lower limits of quantification: retinol 0.01 μM, α‐tocopherol 0.38 μM, β‐tocopherol 0.18 μM, γ‐tocopherol 0.14 μM, and δ‐tocopherol 0.01 μM. Possible application of this method in clinical practice was confirmed by the analysis of human serum samples from healthy volunteers. Finally, the simultaneous determination of retinol and all tocopherol isomers in human serum can enable the clarification of their role in metabolism and in diseases such as cancer.  相似文献

In this study, an enantioselective analytical method based on microwave‐assisted chiral derivatization coupled with ultra high performance liquid chromatography and tandem mass spectrometry was developed for the determination of bambuterol enantiomers in human plasma. The chiral derivatization reaction was greatly accelerated by microwave irradiation. Under the optimized conditions, both the derivatization time and separation time on column was only 3 min, and the lower limit of quantification was 2.5 pg/mL. The recoveries were in the range of 90.1–93.0% without significant matrix effect. Compared with the conventional heating chiral derivatization, microwave‐assisted chiral derivatization obtained higher chiral derivatization yields with much shorter time due to the effect of microwave irradiation. Furthermore, the racemization during the derivatization reaction was systematically investigated. The results showed the concentration of acetic acid and the reaction time had significant effects on the racemization, which could be well controlled during microwave‐assisted chiral derivatization for the short reaction time. Finally, this novel approach was demonstrated by determining bambuterol in human plasma of a clinical pharmacokinetic study in eight healthy volunteers. On the basis of the results, microwave‐assisted chiral derivatization coupled with ultra high performance liquid chromatography and tandem mass spectrometry as a simple and effective enantioselective analysis technique for the determination of chiral drugs in complex biological samples showed great promise.  相似文献

Xiao‐Qing‐Long‐Tang is a traditional Chinese formula used for the treatment of cold syndrome, bronchitis, and nasal allergies for thousands of years. However, the in vivo integrated metabolism of its multiple components and the active chemical constituents of Xiao‐Qing‐Long‐Tang remain unknown. In this study, a method using ultra high performance liquid chromatography coupled with quadrupole time‐of‐flight tandem mass spectrometry was established for the detection and identification of the metabolites in human and rat urine after oral administration of Xiao‐Qing‐Long‐Tang. A total of 19 compounds were detected or tentatively identified in human urine samples, including eight prototypes and 11 metabolites. Also, a total of 50 compounds were detected or tentatively identified in rat urine samples, including 15 prototypes and 35 metabolites detected with either a highly sensitive extracted ion chromatogram method or the MSE determination using Mass Fragment software. Our results indicated that phase Ⅱ reactions (e.g. glucuronidation and sulfation) were the main metabolic pathways of flavones, while phase I reactions (e.g. demethylation and hydroxylation) were the major metabolic reaction for alkaloids, lignans, and ginger essential oil. This investigation provided important structural information on the metabolism of Xiao‐Qing‐Long‐Tang and provided evidence to obtain a more comprehensive metabolic profile.  相似文献
